Following up. I pulled up a project that worked a few days ago and it still
works. I can play MIDi and I can make the project play by pressing play on my
Transport. So, I have no idea what is wrong with any of my default templates.
Still hoping for help.

I’ve determined that I've somehow lost MIDi connectivity, I/O in Samplitude
Pro X3 Suite. No idea how. As far as I know, I have the right MIDi options
checked. If anyone plans to be around TT today, let me know so I can check
off other options.
