A new drive letter will show up when you plug the drive in -- once the drive has been formatted. Right-click the My Computer icon, and select Manage. Double-click Disk Management. Find the drive, right-click, and one of the options should be to format. Be very sure that you're formatting the correct drive, else you may find yourself with lots of data gone.
