I would check your local DNS settings on your Exchange 5.5 server. If they appear to be in order, then it may call for you to reboot your server. BTW, what version of Windows is your 5.5 server? I would move one mailbox as a test, then try to send and receive email both internally and externally. I ran into a problem recently where the Exchange 5.5 server had a different DNS Suffix than what the Internal DNS namespace was, and was causing MTA Bind issues. Once I changed this in the NIC properties, I still had the same problem. It turned out that the DNS entry for the Exchange 5.5 server was never created, so when I created it I restarted the Exchange 2000 MTA Stacks service, and mail was flowing just fine.
