This is a damn good idea!!!!

It would seem easier for transmissive (like backlit prints), since you
wouldn't have to play with the geometry.
Just mask the light source on the lid so that it illuminates only the
patch width or less (we don't want flare from neighbouring patches).
The hard part would be to place a diffraction grating between the
bottom glass and the lens; there's no place, and also where would the
focus point be?
Maybe by building a glass platform on top of the scanner bed, raised
15cm or so... with the diffraction grating just below such platform,
and projecting onto the scanner's glass bed...

quite interesting, indeed, but absolutely draft of course.
