Chris, I had the same problem before. This is mostly due to a previous setting that is still stored in cache. Have you recently made in changes to the incoming website request authentication? I would recommend that you change the website fully qualified domain name to something else and try again. That is you will need to create a new destination set that will point to your website and then publish the site again. This way ISA will not retrieve the previously stored setting.
