disable desktop antivirus. mcafee corp by default blocks port 25 as raj says
check any desktop firewall settings
if all is good then you should be able to see the traffic in the logs on the ISA box

All other applications are blocked as "mass mailers". I had to add telnet.exe manually to the exception for the telnet a.b.c.d 25 to work.
 
Check if Norton does something similar.

Simple question for the pros...
I'm running ISA 2004.
Does it automatically block a telnet session. I'm having an issue
connecting to this one particular domain with my exchange server and
needed to do a telnet session with it but can't get connected with it
inside the firewall. Port 25 is open on the firewall and from home I can
telnet to my server and the offending server.

Do I have something configured wrong on the firewall?
