You talking something like:

=SUM(A3:E3)

being placed in cell G3? That's all it really amounts to.  Go to G3 and 
type in the above at that location.  Youcan hit F2 while sitting in a cell 
to edit the formula behind the cell.  Even the dollar sign ($) stuff you 
may remember from 1-2-3 for making cell coordinates absolute rather than 
relative still works in Excel.

Way back when the earth was still green I used Lotus 123.... Now I need to 
create a spreadsheet in Excel 2003.

Essentially I need to sum a group of numbers, e.g. a3 thru e3 and then in 
g3 the sum must be subtracted by a constant value....

On top of that as I add a new row the function in g3 must be applied to 
the new row in this case g4....

Like I said I've never had to use Excel so any pointers would be most 
appreciated.
