I am in the process of doing some long-overdue clearing out of a
hard-disc and have discovered that the directory "Lost+Found" now
contains 2,010 files amounting to 82,462,326,784 bytes! They are
all dated 21 April 2011 but I cannot think what happened on that
day to produce this data.

Am I reading that correctly - over 82 billion and you didn't notice?

It is likely a significant amount of the disc's contents have been trashed by a crash. DiscKnight was then used to make a repair, and it is likely the machine then booted as normal, but no manual check has been made to see that everything that should be on the disc, still is.

Can someone explain what has probably happened and, more importantly,
say whether I can delete it all to make some very useful space?

I believe they are generated by DiscKnight when correcting errors on
your disc.

Try opening them in a text editor to see what the contents are.

You want him to look at over two thousand files of about 40 Mb each?

The DiscKnight instructions explain exactly what to look for in Lost+Found. But with such a substantial amount of unrecovered data, I would recommend not continuing to use the disc as is, as substantial parts are likely to be missing.

Verify the integrity of any files produced since your last backup, copy them elsewhere, and then reformat and restore from the backup. If, god forbid, you don't have backups, then manually check the directory structure to see what you've lost. If you are very lucky, it may well be the contents of !Scrap or low value data such as an email store.
