Hi milind,
You can certainly configure the users for authentication,
In your ISA server go to servers and Arrays underwhich u can find Your
computer Name(isa server), rigthclick n go to properties , select the
outgoing web requests and select the ask unauthenticated users for
indentifcation check box. Thats it ....

Before which u have to select the users u wish to authenticate, by
creating a protocl rule for HTTP requests and select last tab of this
rule applies to .... Tab and select the users u wish to authenticate....
Note that Your ISA server should have trust with One of the Domain
controllers in your network.
Hope this clarifies.

Vinay,
MCP+I,MCSE.
