You can't. ISA can only do a port to port mapping. Once you point port 80 at DOMAIN1, you cannot also point port 80 at DOMAIN2. You could do port 80 to DOMAIN1 and then say port 81 to DOMAIN2, but then people would have to go to http://www.domain2.com:81 Which I don't think you want to do.
