Well , if I may ask you one more question:
Initially, I had installed ISA on My standalone machine on mY NETWORK and I 
could receive mails but I could not send mails.
I  Could not ping the public IP address on the ISA from the E2K and from the 
domain except from the ISA.

Perfect is one of those goals that is never quite reached. The foundation
and design of the network needs to be carefully planned and laid out.
However, one of the first things that needs to be implemented in a network
that is connected to the Internet is a firewall.

Yes it would expose your mail server until ISA is installed and configured.
That is why the firewall is one of the first to go up. 

I would highly suggest getting Dr. Tom's books and reading them through. 

Again, it can not be stressed enough: If you are going to have publicly
available services (mail server) open to the Internet, a firewall first is a
must.
