From the compiler list, ctrl + shift + F5, choose python, but I don't really use any of the compilation/run functionality as such, from edSharp itsself, but think it changes the context of various other bits of functionality, and alt + shift + I turns indentation mode for jaws etc. on and off.
I primarily use the python interpreter/command line to actually run/test python code that have written using edSharp.

There's a JAWS indentation scheme that comes with it that plays musical notes for each indentation of 2 spaces. You would need to use the JAWS settings packager to install the scheme.
