For Exchange 2K use Exchange System Manager.  Go to Global Settings and Message 
Delivery properties.  Go to the filtering tab and add the domains in this list. 
 You can use wildcards like *@somedomain.com .  Then you have to go the SMTP 
virtual server properies, click Advanced button and check off the Apply Filter 
checkbox.  Cycle SMTP service.

        Speaking of blocking by domain, on Exchange 2000, is there a text list
        of blocked domains that I can edit?  I can edit my Junk Senders list in
        Outlook, but I'd rather filter for everyone at the server level.  It's
        tedious to go through and add domains one at a time, though, so editing
        them in a text format would make it a lot easier.

        I use Exchange Server 5.5 IMS Message filtering to block email by
        domain.
        I, like everyone else, get UCE daily.=20
        One day it will come from: @mail20.C0olmail.com
        Next day it will come from: @mail201.C0olmail.com
        Next day: @mail202.C0olmail.com
        
        And so on.....
        Is there a way to set the blocking in the IMS for all of the
        incarnations of
        this domain (20,201,202,203,305...) in one shot, or do I just have to
        add
        each individually every day?
        Wildcards wouldn't work, would they?
