Connection Timeout is exactly that; ISA timed out trying to reach that site. The error code (10060) is a Winsock error; I have a link to the MSDN Winsock error codes on http://isatools.org. Validate your DNS settings; very often, bad name resolution points you to the wrong server and it's not listening for your request.
