In internet explorer, you'll find the place to enter the proxy details.
I think its under Tool/Internet Options/ Connections tab
 
The proxy address will be the internal nic of you isa

but my the HTTP Redirector Filter is DISABLE 
how can i do my clients as a web proxy clients ? and i wonder if my
clients arent web proxy clients... how cant they access some sites that
incluede destination set..?
