You shouldn't have to.  You may have to restart the appropriate services if
you're looking for instant response to your changes, though.  ISA doesn't
change behavior immediately; it may take up to a minute sometimes to see the
change take place.

When I try to publish a web site last time. I think I did everything
right, but it doesn't work until I reboot the ISA server. Now I am
trying to publish Exchange 2000. I doesn't work, but I cannot find
anything wrong. So I just wonder if I should reboot it again? Do I need
to reboot ISA server each time when I change configuration? Thanks.
