For a useful result, yes.
3a) Will I get a warning or error if I have, say, only 20 measurements?
Not really. It may complain for some reason about a really small
number (i.e. if it is missing patches at some primary combinations), but
otherwise it will forge ahead. Don't expect any sort of accuracy
away from those 20 points though!
