Sounds to me the problem is that you have given read/write/modify permission to system.mdw and someone has opened and locked the file in r/w mode thus preventing anyone else from opening the file. You may need to make the file itself read only. Run up system internals filemon and see what file it is causing the problems. FWIW both the "Common Files" and "Microsoft Office" directory trees are read only for users on our MF servers and Access 2000 works fine ....
