In the Conditional Formatting box are you choosing "Cell Value Is" or "Formula Is"...since you say you are using the > symbol, I'm assuming you are trying to use a formula? Just select "Cell Value Is" and choose "greater than" from the dropdown, then type 100 in the last box.
