Yes it is posssible ....... When you will go through the " Mail Store Create " Wizard it will ask for the database location and there you can give the path whatever you like to set for this Store.

Dear John,
 
It is possible. You can Simply right click on your existing Storage Group it will give you option to create New Store. It will open New Store Wizard which will help you out to create new store.
