If you had the ability to test it on a non-production box I would try it first without uninstalling the SP. That's a good question. I don't have any experience with backoffice installations. According to Microsoft all you need to do is run setup from the enterprise CD. I would make sure that you have a solid backup in place just in case. I did it on a non-production Exchange 2000 standard server and had no issues. If you are thinking about applying Exchange SP2 I would do that after you run the Enterprise installation.
