You should figure on 4-5 seconds scanning time per row to avoid any misreads. I can scan four letter size pages (1848 patches) in 15-20 minutes. The biggest issue is as Matthieu states, positioning the ruler but once you get into the grove it goes smoothly. I would only consider a iSis if you do lots of profiles or run a profiling business.
