You can't do a file based backup of active directory... Do a "System State" backup on your domain controllers, and you will have a backup of the active directory as well. If you are using a third party backup application to backup, and it doesn't support System State backups, either upgrade it, or use NTBackup to backup System State to file, and then backup that file.
