Well, first thing to check is if ISA is stopping it, have a look through the logs in program files\ MS ISA Server\ISALogs and see if its just getting blocked. You could also try installing the firewall client on the machine you're using to see if that allows it. Also, is the connection trying to use a proxy server? I've never had much (read: any) success with that!
