Not if it's only the screw.  You do not have to remove the rocker shaft just to 
replace the adjusting screw.  It's an easy job, just screw in a new one.

Is it the adjustment screw or the rocker arm itself that's stripped?  If it's 
the screw you can replace just the screw.  If it's the rocker arm your gonna 
have to replace the rocker.  I think either way the  rocker arm assy/shaft has 
to come off to accomplish this.
