The only place I have used 2.5D extractors is when I am extracting parasitic information for placed and routed ASICs. Generally this is only distribute RC parasitics. These parasitics are then back annotated into the verilog and static timing analysis is performed. The advent of 2.5D was to address the need for accurate parasitic extraction for very large ASICs, but not take for ever as a 3D solver would.
