I have the wpad and automatic firewall discovery setting available.

The only time the client can get that information from ISA is if it asks
for it.
This process is known as "WPAD".
The ISA help and several articles on isaserver.org cover this in detail.
The best thing to do for tracking down apparent slowness is:
1. leave the settings alone
2. review the ISA logs for connection failures
3. get and review a network capture
