If only having one IIS server, does the second TS server need to be S-Nat
client??

Well, let's be specific.  They won't use a browser to access the terminal
servers- they will use a browser to load the ActiveX control, which will be
used to directly connect to the terminal servers.  I know you know that, but
the OP may not.   Many people, for whatever reason, think the web server
that is providing the ActiveX control actually proxies for the terminal
server.

To that extent, no, 2 installs will not be necessary- one install can
support any number of servers-- you just have to change the .asp page (or
make many pages) to contain the connection information.  And of course, the
Terminal Servers do not have to be the machines that have IIS on them; I
would actually use a box already in the role of a web server rather than
complicate the installations of the other two.
