This is a SMTP config question - ISA isnt where you are looking to set restrictions, but within the SMTP config itself. Use the IIS MMC, select your SMTP virtual server, then right-click ad select properties, then select the access tab. You can allow or restrict whoever you like to either have or not have access by IP or otherwise.
