Fixed the problem - quite a stupid one really! If you goto http://www.sitename.com/exchange it wasn't working, I tried going to http://www.sitename.com/exchange/default.asp and it worked. I just added a few rules to the destination set and everything was up and running. Thanks to everyone who helped. I get 404 file not found when access the site externally, if I create an exchange directory with a default.htm in on the isa server it displays that. So hence it never redirects to the exchange server. I have removed the rule for /* and it gives me 403 Forbidden, the server denies the specified URL.
