Internal Website Error: Connection Error, connection forbidden(10062)

I may not know enough about something here! But it seems odd that, if I
un-configure the the IE proxy settings the whole thing works fine. I
originally suspected the Web Proxy Service on ISA. Your thoughts?

Your website is rejecting the connection from the ISA.
You need to examine the web server logs to determine why.
