Exchange 2000 on the ISA machine is not a good idea, it exposes the AD to the 
internet more easily
than if it was on another server.
I also think it create a pile of headaches for set-up of both components, the 
exchange server and
the ISA server.
Your exchange server will not function properly if there is no domain or dns 
records for that
domain. If other mail servers cannot verify the domain then they reject the 
mail.
