Yes, you can.
The bigger question is this; can you guarantee that the client application will 
always use a specific source port?  If not, such a policy will only cause you 
pain.

Would it be possible to create a custom UDP protocol with a certain port
range and use it in an access rule to allow traffic from certain source ports
(the port range) instead of destination ports?
