Have you installed ISA SP1? There's a fix for that kind of issue, but you'll
also have to set a registry entry as in

  http://www.ISAserver.org

I want to allow only specific sites to be accessed by particular user. I have
created destination set and denied all other destination except the one created
in site and content rule.
ISA doesn't allow access to any other site. When i try to browse the site in
destination the site comes but ISA keeps on asking for passwd again and again
multiple times. When i try to goto next link from that page again same problem.
  Can anyone help me out?

    Hello,

I think, you don't need to use NT backup .

You can back up the ISA server by ISA Management console.
Run ISA management console, expand "server and array" and select the server
which you want to backup and right click the mouse ,point to "backup".
