Have you checked the following:
Does the message specify which server is acting slow for these users?
Do you have the bandwidth?  I realize it's a 5mb link, but is it saturated
or high-latency?
What's a tracert return?
What transports are bound for the Exchange client?
What is the OS type under the clients?

Users in are having periodic issues connecting to the mail server. Many
people are getting an error that says that the Mail Server is unreachable.
They then have to click on retry up to 4 times before they finally connect
and get their e-mail.

Client is outlook 2000/2002 ,accessing remotely with a 5MB link. Server is
Exchange 2000.
