if you want the policy to apply to the entire domain right click the domain
in AD Users & Comps and link the policy you have created to that. if you
want it to apply to selected users (for example everyone but admins) you can
either use the deny option in the security settings of the policy for the
persons or group you want excluded or you can create a new organisational
unit, move the users/groups to be affected into the OU and then apply the
GPO to the OU 'container'.
you can also set things like policy inherentance so that you have policies
at your site or domain level which apply settings everyone should receive
with other policies at the OU level for different groups in your domain.
