Yes and Yes

1. Multiple domains can be hosted by a single Exchange implementation:

2. Best thing would be to create a mail-enabled Public Folder:

Hi, I'm a newbie.

My company has three domains, lets say ABC.co.uk, DEF.co.uk and GHI.com.
Can exchange be set up to handle all three domains?

Also, if I send to sales@xxxxxxxxx can I set exchange up so that only
members of sales within abc get the message and not all my employees?
