Firstly its not a Virus.

DO NOT REMOVE it under any circumstances, unless you want a broken 
computer!!

It is connected with the Windows Registry.

Some processes will lock the file because they are updating or doing 
something with the registry (this can include some anti-virus 
programs).

Do a Google on NTUSER.DAT and you will get lots and lots of info.
