I believe it works in layers, at the bottom is the colour definition of overprint and on top of it is the object layer. However higher levels can't turn off overprint.

If a colour is set to overprint then it overprints where ever it is used, even if the object overprint setting is off.

When trying to set overprint off/on in my TestOPW document, what I noticed was ... I set overprint to one state over the right-hand picture frame, and then moved to the left-hand picture frame to set overprint to the opposite state. On going back to check the overprint setting over the right-hand frame, I found it had changed! Is one of the settings (colour vs object/text level) global in its effects within a document?

Colour definition is global - as described above.

The only explanation why changing the setting for one frame might affect another I can think of, is that both frames were selected at the time.
