Do you know the basic shortcut keys such as r for record, space for stop, etc? Aside from that and understanding how to work with dialogs and menus, you don't need to do anything to improve screen-reader performance. Do you know how to use the Mouse Pointer in Window-eyes to look at different parts of the screen to get information such as the remaining time of an export or when you open a file? Of course, you could create set files which would read you this sort of information but this information is easily found with the virtual mouse and it may well be you won't find the extra convenience of a set file worth the work to create it.

Are you using the beta or nonbeta version of Audacity? The nonbeta version has a minor accessibility problem you need to know about. When you are in a multipage dialog, you will not find the tabs to move from one property sheet to another as you tab through the dialog. You need to use control tab to move among the property sheets. This problem is not present in the beta.
