But  to me "amphibian" means that the bike should
be floatable without further preparations.

I imagine to have a kind of "red button" which automatically inflates the
floaters in a "M" designed James bond gadget vehicle style while going down
the beach and which just start to work wehn hitting the water. 

That would it be! We share the same dream.
(Wasn´t that Q instead of M?!)

another item is the drive: how put the power efficiently to the water for
propulsion?
propellers are for me no option, for I often go to flat streams with lots of
weeds.
Wheels like a Missisippi-Steamer?
kind of Water-drive? 

You should always have an emergency canoe paddle on board ;-)

The propulsion is not the biggest problem, but the sealing and
the buoyancy (Auftrieb).
