Well Id take a slightly different approach Shannon.  First I'd take a piece of 
scrap wood, give it a quick sanding and coat it.  Then see first hand how 
spills clean up.  It might be easier than you think.  Poly does a pretty good 
job of sealing  Yet honestly if it were me, Id be more worried about the water 
getting under it.

Does the water come in from up through the concrete, drain in or just how?  
Even though you have wood over it, the water that gets to the concrete will get 
to the nailers on which you floor is attached and eventually have a mold 
problem.  Some are toxic and many people are very allergic to them.  Keeping it 
dry is the best preventive.

Now for comfort, they make pads that are designed for standing long periods of 
time.  Lots of people that work on their feet a great deal use them.  Easy to 
go get another if its ruined.

I have never had a great deal good to say about vinyl.  Looks may not be that 
important to you.  Sometimes at seams though glue is an issue.  Getting the old 
stuff up is a pain in the tush and back too.  It's a good option, and looks may 
not matter.

Floor paint may be another option and when it gets stained its easy to just 
paint over.  Id still be more worried about the water than which type of floor.
