Just a thought, but coverage should not be an issue
with that magnification factor - in fact, you should
be able to cover the 8x10 with a 4x5 lens at that
magnification.  If a lens covers 4x5 at infinity, you
should be able to cover 8x10 at 1:1.   Also, I am
assuming that you will be going to minimum aperture to
give you depth of field which exceeds the distance
between a gnat's eyeballs, which again helps with
coverage.  It might be possible to do what you want
with a 150mm or even wider lens.  Only issue is that
you will lose distance between your front optic and
the subject, which might make lighting more difficult.
