Hi Jo Ann.
I had to ask this same question just a couple of weeks ago on my first build
from scratch.
I put the CPU and heatsink/fan combo and also the 2 ram sticks on the mobo,
before I mounted it in the case. Much easier to mount in the case before the
power supply is in and getting in the way
I had the whole machine together in the case and after powering it on and
letting it post for the first time, I put in my WinXP disc and rebooted with
it. I installed the mobo drivers disc and vid card drivers disc AFTER
installing WinXP home and Service Pack 2.
Haven't had any problems yet.
I had a friend help with the wires. His vision and expertise are greater
than mine.

Getting ready to put together my first computer from scratch. That is as
soon as I run and get a heatsink and fan for the CPU which I forgot to order
  Power supply is already in case. Thought I would mount the CPU on the mobo
before I put it in the case.  Then plug in power and other wires (other
first ) to board. Then install and plug in drives, install RAM last. Want to
add video card later, but will use built in to get it up and running. Have
installed drives and cards before, but this is my first mobo. When do I use
the CD that comes with the board? Can I use it before I load WinXP? I'm a
little nervous about the wires.
