Are you using one of those USB hubs? Try having the camera (switched on) 
connected to your computer before booting up. 
 
A card reader is the way to go. My HP P4 machine comes with a 6-in-1 card 
reader which is great!
 
Make sure your camera is switched off before removing your card to prevent 
damage to your data or card.
 
Now, before removing your card from the card reader, you have to "eject" it 
first before removing it. Go to 'my computer' to see the drives and right click 
the icon showing your card, choose "eject" and wait till the OS tells you it's 
safe to remove the hardware. Otherwise you would lose all your data. Same goes 
for your camera if connected.
