In addition to what i wrote in the last answer, it's not necessary to 
scan on every boot. In fact, if your antivirus and antispyware programs 
are kept up to date and provide real-time protection, there is no need 
to ever scan. Should there ever be any infected file on the computer 
that got there before it was identified as malware (i.e. not yet in 
signature file updates), you will be denied access if you later try to 
open or run (install) these files. Real-time protection uses the exact 
same signature files as the scan, so scanning provides no extra 
protection and can slow at least older computers down considerably. In 
any case, you should never run 2 scans simultaneously.

In other words, automatically scheduled scans are (like email scanning) 
part of the bells and whistles antimalware manufacturers include so 
people don't forget to be grateful and/or willing to pay.

I also forgot to mention the need to sometimes run an antitrojan scan 
and an online scan.
