Yes, they are different!

Laptop drives are 2.5". Desktop drives are 3.5". However, if you're
using a Mac mini or some comparable x86 clones these "desktops" will
use 2.5" laptop drives.

Beware. The cables are different. If you're using the ATA/IDE
interface you need different cables for both sizes of drive. You can
get 2.5" to 3.5" adapters (the other way around is usually a
pointless/futile exercise since you can't _fit_ a 3.5" drive into
places where 2.5" drives were supposed to be used)... e.g.
