Its a highly debated subject on this and other lists.
My opinion is that for smaller organizations brick
level backups might be fine, but for larger ones its
not practical. With a proper Disaster Recovery plan
its really not needed as well. Its not feasible for
me to backup up a 40GB IS in a brick level fashion as
it would take up 80GB or more on tape (you lose single
instance storage at that point).
With 25 users, I would shut down exchange and find
your priv, pub and dir.edb files. Copy them someplace
on the network with enough space. That will be your
backup in case the SP goes haywire. With those 3
files (in a properly shut down state, i.e. consistent
state) you can recover to that point in time with
little effort. With Exch 5.5 those three files are
it! Before you make any major changes its a good idea
to take offline backups of those. Make sure you know
the name of your ORG, SITE and SERVER as well, mostly
just the server name, but others are good if you need
to do other things, such as do DR on a directory or
something.
Anyway, good luck and have fun :)
