Thanks, you confirmed my suspicions...  It does seem to be related to traffic 
volume, sometimes it works flawlessly, sometimes you cannot get them to play at 
all.  I'll prune down my rule list a bit and see if that helps, otherwise I 
think they're just out of luck.

What you need to do is check the ISA logs for those requests.
You're right; SpewTube streaming app is pretty crappy and not at all tolerant 
of network latency.
It will also be negatively impacted by any 3rd-party apps you may have shoved 
up ISA's butt (can you tell my experience here is poor?) or perhaps the depth 
of HTTP inspection you're imposing.

You may even have to drag out NetMon and see what happens on the wire while you 
test.

Is there any tweak that can be done to make YouTube videos more responsive 
through ISA2006?

YouTube seems to have a really-really delicate timeout setting, and loves to 
throw up the "Video no longer available" message for every video displayed.  
Hitting refresh a dozen times or so will get it to play, but that get a bit old 
after awhile.  Taking the same computer, and putting on the same network on the 
"other side" of the ISA server allows it to work every time.

And no, they didn't like my answer to just not use that site...
