No, because there's absolutely no way to determine which internal server
it should go to.
You've got a house with one door, and two people named Bob. A
deliveryman knocks on the door, and all he's capable of saying is 'I've
got a package for Bob.'
Which Bob gets it? Answer: Can't tell. You need either one house per
Bob, or you need some way to further differentiate. And most of the
major TCP/IP protocols, as I said, can't differentiate. They have no
way to, no provision for, and no concept of.
With HTTP Host headers, you can further differentiate: "I've got a
package for Bob Smith." But this is done at a level above the network
layer.
In other words, ISA could do it (kludge! Kludge!) if it had a proxy app
for the protocol in question. It can do it with Web because there's a
specfic Web proxy. You can do it with SMTP by using a front end/back
end configuration: One server running in front with routing rules (Oh,
it's going to xyz.com, so I resend it to server 1, ooops this one's
going to xxx.com, so I resend it to server 2.)
But for straight NAT based forwarding of arbitrary ports, it's one
instance of a port per IP address.

Surely it's possible to use two internal ip addresses if you disable
socketpooling? Then you could use Destination sets to re-direct.

Can I publish 2 internal servers using the same TCP port
through ISA?
I have just one IP address assigned to my external NIC.
